Progression-free Survival (PFS) Time, as Assessed by an Independent Review Committee (IRC)
Every 6 weeks starting from the date of randomization until occurrence of PD, assessed up to data-cutoff (904 days)

PFS time was defined as the time in months from the date of randomization until first observation of PD (based on imaging as assessed by IRC), or death due to any cause when death occurs within 60 days after the last tumor assessment or randomization (whichever is later). PD is defined as at least a 20 percent (%) increase in the sum of diameters of target lesions, taking as reference the smallest sum on trial; and/or unequivocal progression of existing non-target lesions and/or the presence of new lesions. The sum must also demonstrate an absolute increase of at least 5 millimeter. PFS was measured using Kaplan-Meier (KM) estimates.

Progression Free Survival (PFS) Time
Baseline up to 333 weeks

PFS was defined as the duration (in months) from randomization until the first progressive disease (PD) observation as assessed by the Independent Review Committee (IRC) according to Response Evaluation Criteria for Solid Tumors (RECIST) version 1.0, or death due to any cause when death occurred within 90 days of randomization or the last tumor assessment, whichever was later. PD was defined as at least a 20% increase in the sum of longest diameter (LD) of the target lesions, taking as references the smallest sum LD since the treatment started (including baseline), or appearance of one or more new lesions, and/or unequivocal progression of existing non-target lesions.

PFS Probability Rate at 9 Months in the Dose Escalation Arm
9 months

A precise estimate (+/- 10%) of the probability of not having progression at 9 months. This measure is an estimation derived from the Kaplan-Meier algorithm and does not represent a dimple percentage of participants.

Confirmed Overall Response Rate (cORR) Based on Local Tumor Assessments
From initiation of treatment to disease progression up to a maximum of 17.6 months.

The confirmed overall response rate (cORR) is the percentage of confirmed responses, defined as complete response (CR) or partial response (PR), as assessed by local radiologist/investigator review based on the Response Evaluation Criteria in Solid Tumors (RECIST 1.1). Per Response Evaluation Criteria In Solid Tumors Criteria (RECIST v1.0) for target lesions and assessed by MRI: Complete Response (CR): Disappearance of all target lesions; Partiel response (PR): At least a 30% decrease in the sum of diameters of target lesions, taking as reference the baseline sum diameters. Overall Response (OR) = CR + PR.

Safety Lead-in Phase (SLI): Incidence of Dose Limiting Toxicities (DLTs)
Cycle 1 (up to 28 days)

DLT rate was estimated based on data from DLT-evaluable participants during the DLT-evaluation period (which is the first 28 days after the first dose of study intervention in the SLI) to assess the safety and tolerability of the doublet arm.

What is cetuximab used for?

Cetuximab is used in oncology for cancers including head and neck carcinoma, colorectal cancer, and breast neoplasm. It is studied in EGFR expressing metastatic colorectal cancer and squamous cell carcinoma of the head and neck. It is also known as cetuximab plus platinum based doublet chemotherapy, cetuximab with cisplatin and vinorelbine, and cetuximab with concomitant boost radiotherapy.

What does cetuximab target?

Cetuximab targets EGFR, the epidermal growth factor receptor. It is a monoclonal antibody that acts as an inhibitor of EGFR. This target is relevant in cancers such as colorectal and head and neck carcinomas where EGFR expression is a factor.

Who makes cetuximab?

Cetuximab is developed by Merck KGaA, which trades under the ticker MKGAF. The company is responsible for the clinical development of this monoclonal antibody across multiple cancer indications.

What phase is cetuximab in?

Cetuximab is in Phase 2 clinical development. It is an investigational agent and has been studied in completed trials, including Phase 2 and Phase 3 studies. The development program includes trials in colorectal, head and neck, and gastric cancers.

What clinical trials is cetuximab in?

Cetuximab has been evaluated in completed trials such as NCT05217069 in RAS wild-type colorectal cancer, NCT02383966 in head and neck cancer, NCT01450319 in refractory colorectal cancer, and NCT01388790 in gastric cancer. These trials are completed and enrolled a total of 2444 participants.
